About Screening for Hemoglobin Disorders
Hemoglobin electrophoresis is a well-established technique commonly used in clinical laboratories to screen for hemoglobinopathies (hemoglobin variants and thalassemias) in patient samples.
Many international organizations such as TIF(1), CDC, and APHL(2) recognize the advantages of Sebia’s capillary electrophoresis (CE) technology, including its high resolution and clear hemoglobin fraction profiles in the screening of hemoglobin disorders.
In addition to capillary electrophoresis, Sebia provides solutions based on gel separation techniques: HYDRAGEL HEMOGLOBIN(E) and HYDRAGEL ACID(E) HEMOGLOBIN(E), which, on HYDRASYS instruments, allow the separation of hemoglobins and the detection of major hemoglobin variants: S or D and C or E, through electrophoresis on alkaline agarose gels.
Acid gel electrophoresis, such as HYDRAGEL ACID(E) HEMOGLOBIN(E), should follow to confirm the identification of hemoglobin variants, mainly to differentiate hemoglobins S from D and E from C.
Gel visualization enables both qualitative and quantitative analysis (HbA and HbA2).
The HYDRAGEL HEMOGLOBIN(E) test is based on the principle of electrophoretic separation on agar gel at alkaline pH (pH 8.5).

The HEMOGLOBIN(E) technique is based on the principle of capillary electrophoresis in free solution. Hemoglobin fractions are separated in silica capillaries according to their specific electrophoretic mobility in an alkaline buffer of a given pH and by a more or less significant electro-osmotic flow.
The hemoglobin fractions are directly detected at the specific absorbance of 415 nm. The resulting profile is divided into 15 zones (12 zones for cord blood). Each zone displays a drop-down library of presumed variants migrating specifically in that zone, with more than 500 variants listed.
A whole blood sample is used and analyzed directly on the capillary instruments.
