About Alpha-1 Antitrypsin (A1AT) Phenotyping Test
The most common method for detecting an A1AT deficiency is protein phenotyping by isoelectric focusing.
The HYDRAGEL 18 A1AT ISOFOCUSING kit, used on the HYDRASYS 2 SCAN FOCUSING instrument, enables qualitative detection of different A1AT phenotypes in human serum.
The HYDRASYS 2 semi-automated instrument performs migration, immunofixation, and direct detection on agarose gels to produce A1AT profiles for qualitative analysis.
Phenotyping results, combined with clinical findings and other laboratory tests, support the diagnosis of alpha-1 antitrypsin deficiency.
The HYDRAGEL 18 A1AT ISOFOCUSING test is based on the principle of isoelectric focusing on agarose gel, followed by immunofixation.
After separating the proteins according to their isoelectric point, the gel is directly incubated with a peroxidase-labeled anti-A1AT antiserum, without the need for membrane transfer.
Then, the gel is treated to remove unbound proteins before the enzymatic reaction.
Finally, the gel is visually evaluated.
Human serum samples can be used with the HYDRAGEL 18 A1AT ISOFOCUSING test.

A1AT controls (MM, MZ, and MS) are included in each test batch to quickly identify M, S, and Z phenotypes.
