About Immunofixation Serum and Urine
Immunofixation is an immunological method used to detect and identify monoclonal proteins in serum and urine. After separation of serum and urinary proteins based on their charge, the gel is incubated with various specific antisera. The gel is then treated to remove excess antisera before the final staining step. The gel is visually interpreted to type all immunoglobulins.
The HYDRAGEL IF test is based on the principle of agarose gel electrophoresis followed by immunofixation.
After separation of serum or urinary proteins based on their charge, the gel is incubated with various specific antisera for the characterization of a monoclonal component involved in the gammopathy.

Sensitivity
The minimum detection limit for monoclonal immunoglobulins is approximately 120 mg/L for gels stained with acid violet and around 250 mg/L for gels stained with amido black.
Sensitivity may vary depending on the mobility of the monoclonal component and the polyclonal background.
An internal sensitivity study demonstrated that a Bence Jones protein can be detected in serum at concentrations of:
20 mg/L with antisera against both free and bound light chains
50 mg/L with antisera against free light chains only
